PDB 初期の登録構造と10万件への道のり
1950年代に、科学者は初めて、X線結晶解析によって、原子レベルで、蛋白質の立体構造を解明し始めました。 ミオグロビン1,2 やヘモグロビン3、 リゾチーム4,5、 リボヌクレアーゼ6,7 などの構造が解明されたことにより、予期せずして、蛋白質の規則性や類似性、また、配列、構造、機能、進化の間の関係についての理解が進みました。 科学の発見と理解に対する大きな可能性が早くに認められ、ノーベル賞も授けられました。8こういった初期の構造が、分子構造生物学という科学的試みの新しい分野の誕生につながりました。
たった7つのエントリー(カルボキシペプチダーゼ、キモトリプシン、シトクロム、ヘモグロビン、乳酸脱水素酵素、スブチリシン、トリプシン阻害剤)9-15から始まって、こういった情報豊かな構造を利用できるように、1971年にPDBアーカイブが設立されました。コンピュータ・ネットワークが事実上存在しない時代に、この分野の先駆者がデータをアーカイブし共有する価値を理解していたのは、先見の明があったことの証拠です。
PDBアーカイブが設立されて以降、10-15年ごとに、アーカイブのサイズはおよそ10倍に増えてきました。公開されたPDBエントリー数は、1982年に100件に達し、1993年に1,000件に達し、2000年に10,000件に達しました。2014年5月に100,000件目の構造が公開されると、過去14年間にアーカイブの約90%が公開されたことになります。
