T細胞受容体(PDB:1tcr)

ウイルス(virus)は日々の生活において我々が直面する主な危機の1つで、我々の持つ免疫機構(immune system)はこれらと戦う強力な手段を持っている。我々の細胞はウイルスに感染すると、ウイルスの小片を細胞表面に提示して助けを求める。このウイルスのペプチドを免疫機構が見つけると、その細胞と中にいるウイルスを素早く殺す。先月、ウイルスに感染した細胞が MHC (Major Histocompatibility Complex、主要組織適合性複合体)を用いどのようにしてウイルスのペプチドを提示するかを見てきた。今月は、ウイルスペプチドを認識するタンパク質であるT細胞受容体(T-cell receptor)をみていくことにしよう。

ウイルスの巡視

細胞を殺すT細胞(T-cell)は体内に感染細胞がないかを巡視する警備部隊である。T細胞の表面にあるT細胞受容体は、MHCの中に提示されたウイルスペプチドと強力に結合する。各T細胞はそれぞれ固有のT細胞受容体を持っていて、それぞれが自分に合った型のペプチドを認識する。免疫機構はそれぞれ異なるT細胞受容体を持ったさまざまな種類のT細胞を作り、それによって多くの種類の感染から身体を守っている。このそれぞれ異なる型のT細胞はT細胞が成熟するにしたがって作られる。幹細胞(stem cell)がT細胞へと変化するときに遺伝子を入れ替わり、ランダムで固有なT細胞受容体が形成される。そして、胸腺の分泌過程を通して、我々が持つ通常のタンパク質を認識してしまうT細胞は全て破壊され、外来の物質を認識するT細胞だけが残る。

抗体の腕

ここに示したのはT細胞受容体の一例(PDBエントリー 1tcr )で、 抗体 の片腕と似ている。抗体と同様に、T細胞受容体も2本の鎖でできている。結合部位は分子の先端にあって、数個のタンパク質鎖の環によって形成されている。この環の中にあるアミノ酸はT細胞受容体の種類によって非常に異なっているため、この部分は超可変ループ(hypervariable loop)と呼ばれることが多い。各鎖は一方の端が膜の反対側にある部分も含んでおり、これによって受容体は細胞表面に結合している。この結合部分はこの図では模式的に描かれている。なぜならこの部分が結晶構造を解く際に取り除かれたからである。

T細胞とAIDS

免疫機構の重要性は、それがなくなった時を見れば明らかになってくる。HIV(Human Immunodeficiency Virus、ヒト免疫不全ウイルス)はT細胞に感染し、我々をウイルスから守っている免疫機構そのものを攻撃する。治療しないとウイルスはT細胞を着実に攻撃していき、免疫機構は消耗していく。T細胞の数が少なくなりすぎると、感染者のAIDS(Acquired Immune Deficiency Syndrome、後天性免疫不全症候群)の症状は進行する。そうなると様々な感染を受けやすくなる。なぜなら免疫機構はもはや侵入者と戦う十分な強さを持っていないからである。

分子の握手

死の宣告が確かなものであるかを確かめるため、T細胞と感染細胞は念入りに握手する。最初の接触はT細胞受容体とMHCペプチド複合体との間で行われる(ペプチドは濃い赤で示した)。次にCD分子がMHCの他の場所へと結合し、相互作用を強化する。ここでは2種類の相互作用を示した。MHC I型(MHC class I)は我々が持つほとんどの細胞表面にみられるもので、T細胞受容体および細胞を殺すT細胞の上にあるCD8と相互作用する。MHC II型(MHC class II)は危険なタンパク質を見つけるのに特化した細胞上に主としてみられる。II型はT細胞および免疫機構を刺激する他のT細胞上にあるCD4と相互作用する。これらのタンパク質は全て柔軟な鎖とちょうつがいで細胞膜につながっていて、そのためあちこち移動してこの複合体相互作用を形成することできる。なお上図を描くのに用いたPDBエントリー はPDBエントリー 1akj1bd21fyt1jl41wio の5つである。

構造をみる

T細胞受容体・MHC・ペプチド複合体(左:PDB:1bd2、右:PDB:2ckb)

T細胞受容体とMHCペプチド複合体との相互作用を示す構造がいくつかある。ここに示したのはPDBエントリー 1bd22ckb から得られた初期の構造である。T細胞受容体は図の上部に、MHCは下部に、提示ペプチドは中央部に球で示されている。T細胞受容体は、ペプチドおよびMHCタンパク質の上部表面と相互作用する。この2つの構造は似ているが、T細胞受容体は少しずれてそれぞれのペプチドに最適な形になっていることに注目して欲しい。
